Why Do Some Salivary Glands Stop Working Properly?

  1. Oxidative stress can cause salivary glands to fail, especially in people with conditions like Sjögren's syndrome, diabetes, or those who have undergone radiation therapy.

  2. Golnaz Golnarnik's study found that calcium signaling is disrupted when oxidative stress occurs, leading to salivary gland failure.

  3. The parotid gland (near the ears) and submandibular gland (under the jaw) respond differently to oxidative stress.

  4. Oxidative stress can damage essential parts of cells, including those in salivary glands.

Topic: Health

Researchers at the University of Oslo found that oxidative stress can cause salivary glands to fail. This is especially true for people with conditions like Sjögren's syndrome, diabetes, or those who have undergone radiation therapy. The study suggests that this may be a major reason why some people experience dry mouth and difficulty swallowing.

For people with certain health conditions, such as cancer patients undergoing head and neck radiation therapy, Sjögren's syndrome, diabetes, or older adults, reduced saliva production can have serious consequences. It can make eating, speaking, and swallowing difficult, alter taste perception, and increase the risk of dental cavities and oral infections.

Researchers at the University of Oslo wanted to understand what happens inside salivary glands when they stop working properly. Golnaz Golnarnik, a researcher in the Department of Oral Biology, Faculty of Dentistry, University of Oslo, investigated this question in her doctoral research. She found that oxidative stress is a major contributor to salivary gland failure.

Oxidative stress occurs when a cell's natural defense system becomes overwhelmed. This can happen when cells are exposed to stressors like radiation or inflammation. Normally, our cells have built-in antioxidant systems that help neutralize reactive oxygen species and keep cells safe. However, when this balance is disrupted, oxidative stress can damage essential parts of the cell.

Golnaz Golnarnik's study focused on how calcium signaling inside cells is affected by oxidative stress. She found that when oxidative stress occurs, it disrupts the normal flow of calcium signals that tell cells to produce and secrete saliva. This can cause salivary glands to stop working properly.

The study compared two main salivary glands: the parotid gland (near the ears) and the submandibular gland (under the jaw). Golnarnik found that both glands responded differently to oxidative stress, with the parotid gland being more vulnerable. She also observed changes in metabolism and calcium signaling in parotid gland acinar cells.

This research can help us better understand why some people experience dry mouth and difficulty swallowing. It may also lead to new treatments for conditions like Sjögren's syndrome and diabetes.

Why It Matters

Understanding salivary gland failure is crucial for developing new treatments for conditions like Sjögren's syndrome, diabetes, and radiation therapy side effects. This research can improve the quality of life for millions of people worldwide.
